metadata.dc.description.resumo: | A batata-doce é uma cultura rústica, de ampla adaptação e com grande conteúdo de amido, sendo assim com inúmeras aptidões. Para transformação da batata-doce em biocombustível, alguns processos são necessários. O sucesso da produção de etanol a partir da batata-doce depende da transformação do amido em açúcares fermentescíveis. O processo de conversão mais utilizado é a hidrólise enzimática, a qual utiliza enzimas comercialmente disponíveis. Porém, este processo pode ser realizado através de enzimas secretadas por fungos filamentosos. Neste trabalho foi avaliado o potencial amilásico de fungos filamentosos que colonizam naturalmente a batata-doce. Estes foram isolados, testados quanto à produção enzimática e identificados morfologicamente. Foram isolados 51 fungos da polpa das batatas- doces Beauregard e BRS-Amélia. Deste total, 27 linhagens apresentaram capacidade de hidrólise do amido através da fermentação em estado sólido. Destes, seis revelaram índice enzimático ≥ 2,0 e foram identificados como Aspergillus sp e Penicilium sp. O extrato bruto do gênero Aspergillus sp., mostrou os maiores resultados na conversão em glicose. Este fato indica o potencial destas linhgens na produção de amilase, importante enzima para a produção de etanol. |
Abstract: | The sweet potato is a rustic culture, widely adapted and with a high starch content, thus having innumerable aptitudes. For the transformation of sweet potatoes into biofuel, some processes are necessary. The success of ethanol production from sweet potatoes depends on the transformation of starch in fermentable sugars. The most used conversion process is enzymatic hydrolysis, which uses commercially available enzymes. However, this process can be carried out through enzymes secreted by filamentous fungi. In this work, filamentous fungi that naturally colonized sweet potatoes were studied. These were isolated, tested for enzymatic activity and identified by microculture. 51 fungi from the pulp of sweet potatoes Beauregard and BRS-Amélia were isolated, 27 of them showing the ability to hydrolyze starch. Of these, six showed an enzyme index ≥ 2.0 and were identified as Aspergillus sp and Penicilium sp. The crude extract of the genus Aspergillus sp., showed the greatest results in the conversion to glucose. This fact indicates the potential of these fungi in the production of amylase, an important enzyme for the production of ethanol. |
