metadata.dc.description.resumo: O propósito deste estudo foi avaliar “in vitro” o efeito da desproteinização dentinária mediante a aplicação do hipoclorito de sódio 5,2% em dentina erosionada na resistência de união, nanoinfiltração e morfologia da camada híbrida, utilizando três diferentes sistemas adesivos. A amostra consistiu em 45 molares humanos, cujas superfícies oclusais foram expostas até a exposição da dentina média. Após padronização da smear layer e indução da erosão, os dentes foram divididos de forma aleatória em 3 grupos de 15 dentes de acordo com o sistema adesivo utilizado: Grupo GS1 - Adper Single Bond™ 2/3M-ESPE™, sistema adesivo simplificado de 2 passos; Grupo GS2 - AdheSE® Ivoclar/Vivadent, sistema adesivo autocondicionante de 2 passos e Grupo GS3 – Single Bond Universal™/3M-ESPE™, sistema adesivo autocondicionante de 1 passo. Antes da aplicação do sistema adesivo, cada grupo foi dividido em 3 subgrupos com 5 dentes, de acordo com o tratamento ao qual a dentina foi submetida: A1 - Dentina hígida (controle), A2 - Dentina erosionada e A3 – Dentina erosionada + desproteinizada com NaOCl (5,2%) por 40 seg. Após a aplicação e a fotoativação dos sistemas adesivos, de acordo com as especificações do fabricante, os dentes foram restaurados com resina composta e seccionados para obtenção de espécimes em forma de “palitos” (0,8 mm2). O teste de resistência de união (RU) foi realizado após 24h de armazenamento em água a 37 ºC a uma velocidade de 1 mm / mim. Antes do teste de microtração, foram separados aleatoriamente 4 “palitos” de cada condição experimental para análise da formação de tags e nanoinfiltração (NI) em MEV. Os dados de RU e NI foram analisados por ANOVA de 2 fatores complementado pelo teste de Tukey (α = 0,05). O sistema adesivo autocondicionante Single Bond Universal após desproteinização apresentou a maior média de RU (41,86 ± 5,80 MPa). A menor média de RU foi observada para o sistema adesivo autocondicionante AdheSE® após erosão sem desproteinização (14,04 ± 7,28 MPa). A maior média de NI (%) foi observada para o sistema adesivo autocondicionante AdheSE® após erosionado sem desproteinização (69,40 ± 8,73) e o sistema adesivo Single Bond Universal apresentou a menor média de NI (%) para o grupo desproteinizado (14,48 ± 5,47). A observação das fotomicrografias para dentina hígida e desproteinizada em relação a todos os sistemas adesivos notou-se a presença de tags de maior comprimento na camada híbrida. A desproteinização dentinária acarretou um aumento de RU no substrato erosionado para os grupos em que foram aplicados sistemas adesivos autocondicionantes. Em relação à NI, melhores condições foram obtidas para o sistema adesivo simplificado Universal.
Abstract: The purpose of this "in vitro" study was to evaluate the effect of the deproteinization of the eroded dentin in bond strength, nanoleakage and morphology of the hybrid layer using three different adhesive systems. The sample consisted of 45 human molars. The teeth had their occlusal surfaces exposed to mid coronal dentin. After the standardization of smear layer and the induction of the dentin erosion, they were randomly divided into 3 groups (n=15), according to the adhesive system used: GS1 - Adper Single Bond™ 2 (3M-ESPE™) - 2 steps simplified adhesive system; GS2- AdheSE® (Ivoclar/Vivadent) - 2 steps self-etch adhesive system; GS3- Single Bond Universal™ (3M-ESPE) - 1 step adhesive system. Previously to the application of the adhesive system, each group was divided into 3 subgroups (n=5), according to the treatment of the dentin: A1– Sound Dentin (control), A2- eroded dentine and A3- eroded dentine + deproteinized with NaOCl ( 5.2% ) for 40 sec. After the application of the adhesive systems according to the manufacturer's instructions, the teeth were restored with composite resin and sectioned to obtain “sticks-like” resin specimens (0.8 mm2). The microtensile bond strength test (TBS) was performed after 24h storage in water at 37 ºC at a rate of 1 mm/min. Before the microtensile test, 4 “sticks” of each experimental condition were randomly assigned for micromorphological analysis (formation of the resin tags and nanoleakage - NI) by SEM. The TBS and NI data were analyzed by 2 way ANOVA complemented by Tukey's test (α=0.05). The group treated with Single Bond Universal and deproteinization showed the highest μ-TBS (41.86 ± 5.80 MPa). The lowest μ-TBS mean was observed for AdheSE® after erosion without deproteinization (14.04 ± 7.28 MPa). The higher NI mean (%) was observed for AdheSE® after erosion without deproteinization (69.40 ± 8.73) and Single Bond Universal had the lowest NI mean (%) for the deproteinized group (14.48 ± 5.47). The photomicrographs of sound dentin and deproteinized dentin for all adhesive systems showed the presence of longer resin tags in the hybrid layer compared to the eroded dentin only The dentin deproteinization resulted in increased TBS in the eroded substrate for the groups in which self-etch adhesive systems were applied.. For the NI, the best conditions were obtained for the Single Bond Universal adhesive system.
