dc.description.abstractBACKGROUND: Once the majority of human breast cancers have shown overexpression of insulin receptors, glycose levels and insulin intake may possibly affect the breast cancer outcomes. However, the exact role high-fat diet, hyperglycemia and hyperinsulinemia in breast cancer outcome remains unclear. Thus, the aim of present study was to evaluate the role of high-fat diet, glycemia and insulin intake on murine breast adenocarcinoma tumor growth in vivo. METHODS: Adult female mice (Rodentia muridae, Mus musculus, Balb-C lineage) were submitted to subcutaneous implantation of murine breast adenocarcinoma cells, divided in four groups. Group 1 received only tumor implantation (4T1 cells), Group 2, tumor implantation and high-fat diet (4T1-HFD), Group 3, tumor implantation and insulin (4T1-INS) and Group 4 tumor implantation and high-fat diet (4T1-HFD-INS). Measurements and laboratory tests were taken at days 15 and 30 of experimentation. RESULTS: This paper presents macroscopic findings and laboratory tests. High fat diet is correlated to bigger tumor volume (p=0.0492), except if associated to insulin (0.0168), but not to higher glucose serum levels. Groups that received insulin showed smaller tumor growth amplitude, but there was no difference in glucose serum levels between any group at D30. Serum glucose levels were not correlated with higher tumoral volumes. Tumor invasion of upper left limb was incident in all groups (66.7% to 100%), with no difference between groups (p=0.9471). Pulmonary metastases were more frequent in groups that received insulin (p<0.05) or HFD plus insulin (p<0.05) when compared to group 1 (4T1).pt_BR
dc.description.resumoCENÁRIO: Como a maioria dos cânceres de mama humanos têm demonstrado superexpressão de receptores de insulina, níveis de glicose e uso de insulina podem possivelmente afetar os desfechos clínicos do câncer de mama. No entanto, o exato papel de dieta Hiperlipídica (HFD), hiperglicemia e hiperinsulinemia no câncer de mama permanecem pouco claros. OBJETIVO: avaliar o papel de dieta hiperlipídica, glicemia e uso de insulina no crescimento tumoral de adenocarcinoma mamário in vivo. MÉTODO: Camundongos adultos fêmeas (Rodentia muridae, Mus musculus, Balb-C lineage) foram submetidas a implantação subcutânea de células de adenocarcinoma mamário murino (células da linhagem 4T1), randomizados em quatro grupos. Grupo 1 recebeu somente implantação tumoral (4T1), Grupo 2, implantação tumoral e dieta hiperlipídica (4T1-HFD), Grupo 3, implantação tumoral e insulina (4T1-INS) e Grupo 4, implantação tumoral, dieta hiperlipídica e insulina (4T1-HFD-INS). Medidas e testes laboratoriais foram aferidos nos dias 15 e 30 de experimentação. RESULTADOS: este artigo apresenta resultados macroscópicos e laboratoriais. HFD esteve correlacionada com maior volume tumoral (p=0,0492), exceto se associada a insulina (p=0,0168), mas não a níveis mais altos de glicemia. Grupos que receberam insulina mostraram menor amplitude de crescimento tumoral, mas não houve diferença entre os níveis finais de glicose (no D30) entre os grupos. Os níveis de glicemia não se correlacionaram com maiores volumes tumorais. Invasão tumoral do membro superior homolateral foi comum em todos os grupos (66,7% a 100%), sem diferença entre os grupos (p=0.9471). Metástases pulmonares foram mais frequentes em grupos que receberam insulina (p<0,05) ou HFD e insulina (p<0,05) quando comparados com o grupo 1 (4T1).pt_BR
